Two boys, aged four and 10, and a 32-year-old woman died after the crash on the M6 in Staffordshire at around 11.30am, West Midlands Ambulance Service (WMAS) said.
Two other women were also in the car at the time of the accident and are receiving treatment in hospital.
The tragedy happened on the northbound carriageway near junction 15 at around 11.25am.
A WMAS spokesman said: “Crews arrived to find a single vehicle that had crashed and overturned.
“Unfortunately, despite the best efforts of ambulance crews, nothing could be done to save the two boys and one woman, and they were confirmed dead at the scene.”
